Placental blood flow can influence malaria during pregnancy

Malaria in pregnancy causes a range of adverse effects, including abortions, stillbirths, premature delivery and low infant birth weight. Many of these effects are thought to derive from a placental inflammatory response resulting from interaction of infected red blood cells with the placental tissue. In a study published in the latest issue of the journal PLOS Pathogen*, a researchers' team led by Carlos Penha-Gon?alves at the Instituto Gulbenkian de Ci?ncia (IGC), Portugal, observed, for the first time, the mouse placental circulation and showed how it can influence the malaria parasite behavior and infection. Their results indicate a higher accumulation of parasites in placental regions with low blood flow, being these areas more prone to an inflammatory response.
In humans, red blood cells infected with the malaria parasite, Plasmodium falciparum, accumulate in the placenta via interaction with a molecule expressed on the placental tissue ? a process called sequestration. In response to this event, placental cells secrete substances that recruit inflammatory cells leading to placental damage and negatively impacting fetal growth. Until now placental circulation has not been linked to the infected red blood cell sequestration. In fact, it is not trivial to investigate this hypothesis in human placenta, due to technical constraints
Luciana Moraes, an investigator of Carlos Penha-Gon?alves laboratory, has provided new insights to this issue by developing an experimental system that allowed the live observation of the blood flow in the mouse placenta. Mating two strains of mice, one of them with cells stained with a colorful marker, Luciana was able to identify the placental tissue (fetus origin). In collaboration with Carlos Tadokoro's laboratory at the IGC, the investigators developed a microscopy technique that allowed the observation of the placenta in a living mouse. Immediately before exposure to the microscope the mouse was injected with a fluorescent substance that labels the blood. With this set-up it was possible to distinguish maternal blood and placental tissue. The results showed for the first time how the circulation occurs in the placenta, and that the blood flows with different speeds in different regions of the placenta.
Next, the investigators infected red blood cells with the malaria parasite Plasmodium berghei, stained with a different color, and observed ? live ? the behavior of the parasite inside the placenta. They observed that in the areas with higher blood flow, the parasite never stops moving and does not interact with the placental tissue. The accumulation of parasite just occurs in areas of low or absence of flow. In these regions, placental macrophages engulf the infected red blood cells to attempt parasite clearance. Their observations also suggest that movements of the placental tissue may control the blood flow.
Luciana Moraes says: "Our results indicate that binding of infected red blood cells to a molecule expressed in the placenta may not be the only mechanism of parasite sequestration. The dynamics of placental circulation may also play an important role, and should be considered when designing therapeutics."
Carlos Penha-Gon?alves adds: "This is the first study done that shows live how placental blood circulation impacts on the local infection by the malaria parasite. It would be interesting and worthwhile to explore if a similar process occurs in the placenta of humans, taking in consideration that microcirculation in human placenta is quite different."

Malian military: 4 dead, 5 wounded in mine blast
SEVARE, Mali (AP) ? A Malian military spokesman says four Malian soldiers died and five others were wounded when their vehicle hit a land mine in eastern Mali.
Modibo Traore told The Associated Press on Thursday that the soldiers were killed Wednesday evening in the town of Gossi.
The town is located between the towns of Hombori and Gao, both of which had been under Islamist rule until the French-led military offensive began earlier this month.
The French and Malians have been able to grab back control of the provincial capitals of Timbuktu, Gao and Kidal in a matter of weeks.
However, questions remain about whether the Malian military and its West African allies can hold the cities once the French leave.

Indian SuperMeals: Toddler and Baby Cookbook up for review ? But ...
We have been lucky enough to be asked to try out Zainab Jagot Ahmed?s baby and toddler cookbook, Indian SuperMeals. It is available in ebook form so I have the one that is compatible with my Ipad. Zainab was inspired to write this book when she discovered that there were no baby and toddler cookbooks with Asian or Asian influenced recipes. She was starting the weaning process with her daughter Aaliyah and wanted to?introduce Asian flavours early to both broaden her palate and introduce her to her culinary heritage.?With a lot of research into dietary requirements for weaning babies and toddlers as well as spices and superfoods, Zainab began creating her own dishes and soon had family and friends asking for recipes.
Firstly I was impressed with how well researched this cookbook is, it is packed with nutritional information and top tips to make your baby or toddler?s food not only more full of flavour but also with ways to boost baby?s immune system. Both big pluses in my opinion!
After a comprehensive introduction covering SuperSpices and SuperMeals, the book is then split into the stages of weaning with relevant recipes. Obviously with my girls no longer being babies we focused on the section, ?Family Supermeals Stage 4: 1-3 years?.?There is a lovely range of recipes in this bit and I instantly spotted lots that I wanted to try out. There?s not only curries but also old favourites such as cottage pie with an Indian twist, as well as recipes for rotis and dhal. I have to admit that I am often such a cop out when it comes to curries and often resort to jars or *ahem* takeaways! So the recipes we tried out were primarily curries ? need some good ones to add to my weekly meal plan.
After stocking up my herbs and spices cupboard with a few extra bottles and jars we were ready to go. I found the recipes that we tried simple and easy to follow, so perfect for midweek post school run, post-swimming lesson, etc! There was a mixed reaction to the curries made, with Little E (aged 2) and both myself and my husband loving the food, and 5 year old Noo turning her nose up at curry that wasn?t, and I quote ?yellow?. In other words it wasn?t beloved [take away] Korma! This hasn?t put me off as Noo is notoriously fussy with food and I am determined to keep trying different foods with her as I know that one day she will come round.
I would recommend this cookbook to any families starting the weaning process, not just families with Asian heritage.?I can honestly say that I wish this book had been around when I was weaning both of my girls. I know that Noo would be less fussy now if I had broadened her palate from the start and not just given her the standard and often rather bland weaning foods. I am thankful that Little Miss E enjoys trying new food and hope that this will continue as she gets older, I know that this cook book will definitely help us with that aim. The only thing I would change is that I would love to own it in real hard copy book format. This is because I am one of those?people who misses the feel of a ?real? book when reading from a screen. That is my only negative though!
Over the next two of weeks we will be featuring a couple of recipes from the Stage 4 section of Indian SuperMeals, starting with Lamb, Saag Aloo Curry on Tuesday (5th of February) ! So be sure to keep a look out for them.
